摘要
目的探讨外源性Noggin对肝大部切除大鼠肝再生率的影响。方法80只雄性Wistar大鼠,取72只行肝大部切除术(PH),随机分为生理盐水组(NS)、Noggin低剂量组(NL)和Noggin高剂量组(NH),余8只为正常对照组(NC)。以NC组为0h观察点,在PH后12h、48h、72h测定肝再生率、骨形成蛋白2(BMP2)含量。结果PH后NS、NL、NH组肝再生率逐渐增高,NL、NH组与NS组间在12h差异有统计学意义(P<0.01)。PH后NS、NL、NH组肝组织匀浆BMP2含量在12h降至最低,此时NL、NH组与NS组间差异有统计学意义(P<0.01)。结论外源性Noggin显著抑制肝大部切除大鼠残留肝组织BMP2含量,通过BMP2对肝再生的负性调节作用间接提高肝再生率。
Objective To investigate the effect of exogenous Noggin on the liver regeneration rate after partial hepatectomy in rats. Methods Partial hepatectomy was performed in 72 of 80 Wistar rats and the remaining 8 Wistar rats as normal control group. The 72 rats were divided into three groups: normal sodium group(NS),low close Noggin group(NL) and high dose Noggin group(NH),and the liver regeneration rate and the BMP2 level were measured 12h,48h and 72h after partial hepateetomy,with normal control group as Oh observation time point. Results The liver regeneration rate in all groups was gradually increased after partial hepatectomy. The liver regeneration rate in NL and NH groups was significantly decreased than that in NS group at 12h after partial hepatectomy(P〈0.01 ). The liver homogenate BMP2 level in NS,NL and NH groups was significantly decreased than that in NC group at 12h after paxtial hepatectomy,and the BMP2 level in NL and NH groups was significantly decreased than that in NS group at 12h after partial hepatectomy(P〈0.01). Conclusion Exogenous Noggin can significantly inhibit the BMP2 level in the remnant liver after partial hepatectomy in rats,and indirectly promote liver regeneration rate by down-regnlating the effect of BMP2 on the hver regeneration.
