摘要
采用RT-PCR方法从成年麦洼牦牛脾中克隆到同种移植炎症因子AIF-1的编码基因,并研究了其在各组织中的表达模式。结果表明,AIF-1基因包含一个444 bp的完整阅读框,编码一由147个氨基酸组成的蛋白,推测其分子质量约为16.6 ku,与其他多种动物的AIF-1的氨基酸序列有89%-95%的同源性。RT-PCR半定量分析结果显示,AIF-1基因在麦洼牦牛的脾、肾中都有转录,在脾中表达水平相对较高,而在肝、乳腺、心、肌肉中未见明显表达。
An allograft inflammatory factor I(AIF-1) gene was cloned from the spleen of adult Maiwa yak by using reverse transeription-polymerase chain reaction(RT-PCR), and the expression module of AIG 1 gene in various tissues of yak was observed. The AIF-1 gene was 555 bp,containing a 444 bp open reading frame which encodes a 147 amino acids of a 16. 6 ku protein of molecular weight. The encoded protein shared 89% to 95% identity with other species of animals. Semi-quantitative RT-PCR analyses showed that mRNA expression of Maiwa yak AIF1 gene was high in the spleen and kidney,and the expression level in the spleen was higher than that in the kindey, but could not be detected in the heart,liver, muscle and galactophore.
