急性有机磷中毒小鼠血肌酶水平的变化及其意义 被引量:1

摘要 目的研究急性有机磷中毒(AOPP)小鼠血肌酶水平的变化及其意义。方法将152只鼠龄为8周的雄性昆明小鼠分为正常对照组(8只)、生理盐水对照组(10只)及AOPP组(135只);AOPP组分为染毒后30 min、2 h、4 h、12 h、1 d、2 d、3 d、4 d及7 d亚组并分笼(15只/笼)。腹腔注射氧化乐果(50 mg/kg)和阿托品(10 mg/kg)。在各时间点随机取AOPP组各亚组中的8只小鼠进行血胆碱酯酶(CHE)、血清肌酸激酶(CK)、天门冬氨酸转氨酶(AST)及乳酸脱氢酶(LDH)水平检测,并与正常对照组和生理盐水对照组比较。对染毒后1~4 d肌无力小鼠与肌力正常小鼠的血肌酶水平进行比较。结果与正常对照组及生理盐水对照组比较,AOPP组染毒后30 min^3 d亚组血CHE水平明显降低(均P<0.05),4 d、7 d亚组血CHE水平与正常对照组差异无统计学意义。各时间点亚组血AST、CK、LDH水平明显增高(均P<0.05);AOPP组发生肌无力小鼠的血AST、CK、LDH水平显著高于肌力正常小鼠(均P<0.05),而血CHE水平差异无统计学意义。结论AOPP中毒小鼠的血肌酶水平显著升高,发生肌无力小鼠的血肌酶水平高于肌力正常小鼠。提示血肌酶水平可作为判断中间期肌无力综合征(IMS)发生的监测指标。 Objective To investigate the change and its significance of blood level of myocardium zymogram in acute organophosphrus poisoning(AOPP) mice. Methods 152 eight-week-old male mice were divided into 3 groups randomly : normal control group( n = 8 ) ; normal saline control group ( n = 10 ) ; AOPP group ( n = 135 ). The AOPP group mice were assigned by 9 time-points (30 rain, 2 h, 4 h, 12 h, 1 d, 2 d, 3 d, 4 d and 7 d after poisoning). 15 mice were in per cage. The mice in AOPP group were introperitoneal injected Omethoate (50 mg/kg) and Atropine ( 10 mg/kg). Centhifugalizing the blood and the serum were taken, examined cholineesterase(CHE) , creatine kinase (CK), aspartate aminotransferase(AST) and lactate dehydrogenase(LDH). The level of myocardium zymogram of myasthenia mice was compared with normal muscle mice in the AOPP group. Results Compared with normal control group and normal saline normal saline control group, CHE activity was persistent inhibition in the AOPP 30 min - 3 d subgroups ( all P 〈 0. 05 ) , it was no significant differences of the level of CHE in 4 d and 7 d subgroups. The levels of AST, CK, LDH were significantly higher in each AOPP subgroups ( all P 〈 0. 05 ). The levels of AST, CK, LDH in myasthenia mice were higher than those in the normal muscle mice ( all P 〈 0. 05 ). Conclusions The level of blood myocardium zymogram is significantly higher in AOPP mice. Compared with normal muscle mice, myocardinm zymogram is higher in myasthenia mice. Which indicates that blood level of myocardium zymogram can be the monitor index in judging the occurrence of intermediate myasthenia syndrome.
