摘要
目的观察细胞外基质的受体—整合素α5在胚胎小鼠肾脏发育过程中的时空性表达,探讨其在胚胎肾脏发育中的作用。方法采用免疫组织化学(酶组织化学和荧光组织化学)方法观察不同胚龄小鼠肾组织中整合素α5的表达,细胞图像分析技术检测阳性表达的灰度值,体视学技术检测阳性表达的体密度值。结果免疫组化结果显示:胚龄12 d时整合素α5在输尿管芽(UB)周围的生后肾组织有表达。胚龄14 d时整合素α5在生肾区有表达。胚龄16 d和18 d时整合素α5在发育各个阶段的肾小体、肾小管都有一定程度的表达。图像分析和体视学结果显示:整合素α5的表达随着肾脏发育逐渐增强。结论整合素α5在发育各个时期的肾小体、肾小管都有一定程度的表达,且随着肾脏的发育表达逐渐增多。提示整合素α5对肾小体、肾小管的发育都起一定的诱导作用。
: Objective To observe the chronological and spatial expression of integrin α5 , which is the receptor of extraeellular matrix (ECM) in developing mouse kidney of mouse, in order to clarify its function during nephrogeneisis. Methods The expression of integrin ots was examined by immunohistoehem- istry (IHC, enzymatic and immunofluorescence histochemistry) , analyzed by computerized densitometry and stereology. Results Integrin α5 was expressed in the metauephrogenic tissue near ureteric bud (UB) on embryonic day 12 (ED 12). It was expressed in the nephrogenic zone on ED 14. On both ED 16 and ED 18, the expression of integrin α5 was seen in both glomeruli and tubules. The computerized densitometry and stereology revealed that the expression of intergrin α5 was enhanced with the development of kidney. Conclusion The expression of intergrin α5 can be seen in both glomeruli and tubules in the developing kidney,and its expression is enhanced with the kidney development. It suggests that intergrin α5 plays an most important role in the kidney development.
