联合抗氧化微量营养素对小鼠胰岛β细胞保护作用的超微结构与形态计量学研究

摘要 目的电镜观察及定量分析联合抗氧化微量营养素(combined antioxidant micronutrients,CAM)对胰岛β细胞保护作用。方法采用小剂量多次注射链脲佐菌素(muitipe low dosage ofstreptozotocin,MLDS)方法制备T1DM(type 1 diabetes mellitus)小鼠模型,分别添加4种AM和7种AM干预,应用透射电镜摄片,形态学定量评价胰岛β细胞超微结构变化。结果微量营养素添加组小鼠血糖水平明显低于糖尿病模型组;胰岛β细胞内分泌颗粒数量明显高于糖尿病模型组(p<0.01);胰岛β细胞内分泌颗粒核芯与界膜之间空隙明显大于糖尿病模型组(p<0.01)。结论联合应用抗氧化微量营养素对T1DM模型小鼠胰岛β细胞超微结构具有保护作用。 Objective To observe and analyze the effects of combined antioxidant micronutrients (CAM) on the uhrastructure of pancreatic β cells in mice with type I diabetes mellitus (TIDM). Methods The TIDM mouse model (sixty male C57BL mice, body weight 22 ±2 g )was prepared by multiple injection of low dose streptozotocin (MLDS). Forty diabetic mice supplemented with CAM were divided into two groups: the CAM4 group (selenium, vanadium, chromium and vitamin E) and CAM7 group (selenium, vanadium, chromium, Vitamin E, Vitamin C, lipoie acid and niacinamide) , 20 mice in each group. The uhrastructural observation of the pancreatic islets in mice of each group was conducted by transmission electron microscopy, and the electron microscopic photographs were measured by a computer image analysis system. Results The blood glucose levels in the mice of groups supplemented with combined micronutrients were significantly lower than those of TIDM model group. The endocrine granules in β cells were significantly more abundant and the distance between the cores of endocrine granules and limiting membrane was significantly longer in both CAM4 and CAM7 groups. Conclusion Combined antioxidant micronutrients may exert a protect effect on the ultrastrueture of pancreatic β cells in TIDM mice.
