摘要
目的:探讨痹祺胶囊对骨性关节炎关节软骨破坏的干预作用。方法:6月龄新西兰大耳白兔25只,随机分为假手术组、模型组、痹祺胶囊低剂量组、痹祺胶囊中剂量组、痹祺胶囊高剂量组,每组各5只。使用ELISA方法测定血清MMP-3、TIMP-1及IL-6含量,HE染色进行关节软骨组织形态学观察。结果:组织形态学观察痹祺胶囊可改善骨性关节炎的软骨病理改变。中、高剂量痹祺胶囊组血清中的MMP-3、IL-6含量明显低于模型组(P<0.05),TIMP-1/MMP-3较模型组升高。结论:痹祺胶囊对骨性关节炎模型关节软骨破坏具有一定的保护作用,其机理可能与降低血清中MMP-3、IL-6含量,改善TIMP-1/MMP-3水平有关。
Objective To investigate the chondroprotective effect of Biqi(痹祺) capsule on articular cartilage of rabbit knee joint with osteoarthritis. Methods Twenty-five New Zealand rabbits were randomly divided into 5 groups:sham group,model group,low-dosage,middle dosage and high-dosage groups of Biqi capsule.Eight weeks after the operation,the concentration of matrix metalloproteinase-3 (MMP-3),tissue inhibitors of metalloproteinase-1 (TIMP-1) and interleukin-6 (IL-6) in serum was determined by ELISA,respectively;the articular cartilage morphological changes in every group were observed by section stained with HE. Results The morphological condition of osteoarthritic articular cartilage could be improved in the three Biqi capsule groups by histological and gross morphologic observation.The concentrations of MMP-3 and IL-6 in the middle-dosage and high-dosage group of Biqi capsule serum were significantly lower than those of the model group (P〈0.05),and the ratio of TIMP-1/MMP-3 was improved compared with the model group also. Conclusion Biqi capsule may offer some protective effect on articular cartilage of rabbit knee joint with osteoarthritis,this may attribute to the reduction of the concentrations of MMP-3 and IL-6 and the improvement of ratio TIPM/MMP-3.
