摘要
目的探讨过敏性紫癜患儿外周血单个核细胞(PBMC)中CD40LmRNA的表达及其与血清中IL-6和IL-8的关系。方法应用逆转录-聚合酶链式反应(RT-PCR),从转录水平检测30例过敏性紫癜(HSP)患儿和20例正常儿童(对照组)PBMC中CD40LmRNA的表达,并应用ELISA双抗体夹心法检测血清中细胞因子IL-6和IL-8的水平。结果HSP患儿(PBMC)中CD40LmRNA的表达明显高于对照组(P<0.05);HSP患儿血清中IL-6和IL-8水平明显高于对照组(P<0.05),加入抗-CD40LmAb后恢复正常。结论HSP患儿PBMC中CD40L的表达异常增高,且CD40L的表达与血清中IL-6和IL-8呈正相关。
Objective To investigate the expression of costimulatory molecules CD40L mRNA in patients with Henoch-Schonlein purpura ( HSP) and explore the relationship between CD40L mRNA and cytokine IL-6, IL-8 in serum. Methods Thirty HSP patients and twenty healthy children were recruited. The expression of CD40L mRNA on peripheral blood mononuclear cells ( PBMC) were quantitatively analyzed by reverse transcription-polymerase chain reaction (RT-PCR) technique and the level of cytokine IL-6 and IL-8 were measured by ELISA. Results Compared with normal controls , the expression of CD40L mRNA in PBMC significantly increased in patients with HSP (P 0.05). The expression of IL-6 and IL-8 in serum of patients with HSP were significantly higher than the controls (P 0.05), and decreased to normal level after adding antiCD40L mAb. Conclusions The abnormally high expression of CD40L may play a role in pathological process of HSP. The expression of CD40L has positive correlation with serum IL-6 and IL-8.
