摘要
基底的物理硬度和表面形貌对锚着细胞的生物学行为有着深远的影响。生长在不同硬度基底的细胞其形态特征、粘着斑装配、细胞骨架状态都有很大差别。此外硬度梯度和各向异性可以诱导细胞定向迁移和生长,这些行为在许多病理生理过程中起到关键性作用,如组织发育、纤维化病变、肿瘤转移等。本文总结了近些年来有关方面的研究进展,重点讨论了基底硬度在细胞表型、组织稳定性维持方面的作用,并且对有关的信号转导机制进行了概述。
Mechanical rigidity and surface morphology of the substrate have great impact on the biological behavior of the anchoring cell. Cell morphology, focal adhesion assembly as well as cystoskeleton organization have been shown to differ when cells were cultured on substrate with different stiffness. Furthermore, anisotropic substrates with stiffness gradients induced directed migration of the cells. These behaviors may play important roles in physiological and pathologic processes, such as tissue development, fibrosis and tumor metastasis. This review summarizes the progresses on this topic during the past few years. Focus was placed on the role of substrate rigidity on the maintenance of cell phenotype and tissue organization, as well as the relevant signaling mechanisms.
