摘要
研究复合成骨蛋白-1(osteogenic protein-1,OP-1)多肽水凝胶缓释系统对小鼠成骨细胞碱性磷酸酶(alkaline phosphatase,ALP)、骨钙素(osteocalcin,OC)和核心结合因子α1(corebinding factor α1,Cbfα1)表达的影响。实验分OP-1组、OP-1/水凝胶组和对照组,对小鼠成骨细胞培养1d、4d、7d、10d、14d后,运用免疫细胞化学方法分别检测各组各时间点ALP、OC和Cbfαl的表达。结果显示培养4d、7d、10d、14d,OP-1组和OP-1/水凝胶组ALP、OC、Cbfαl阳性表达均比对照组多(P<0.05),培养4d、7d、10d,OP-1/水凝胶组ALP、OC、Cbfαl阳性表达均较OP-1组少(P<0.05);OP-1组培养1d,Cbfαl开始有表达,而其他组、其他指标在1d均无表达;OP-1组培养7d,ALP和Cbfαl表达量达顶峰,而OP-1/水凝胶组培养10d才达顶峰;到14dOP-1组和OP-1/水凝胶组ALP、Cbfαl的表达无显著性差异(P>0.05)。研究表明复合OP-1多肽水凝胶缓释系统能缓慢释放OP-1,促进成骨细胞分化,增加ALP、OC和Cbfαl的表达。
Study on the effect of the polypeptide hydrogel delivery system combined with osteogenic protein-1 (OP-1) on expression of alkaline phosphatase (ALP), osteocalcin (OC) and core binding factor α1 (Cbfα1) in mouse osteoblasts. Culture cells and divide them into three groups (OP-1 group, OP-1/hydrogel group and control group). The expression of ALP, OC and Cbfα1 in these cells were observed with irnmunocytochemistry after culturing for one day, four days, seven days, ten days and fourteen days. The expression of ALP, OC and Cbfctl of OP-1 group and OP-1/hydrogel group were all more than that of control group after being cultured for four days, seven days, ten days and fourteen days (P〈0.05). The expression of ALP, OC and Cbfα1 of OP-1/hydrogel group were fewer than that of OP-1 group after being cultured for four days, seven days and ten days (P〈0.05). There was a little of expression of Cbfα1 of OP-1 group after being cultured for one day. But there was none of the others. The expression of ALP and Cbfα1 of OP-1 group reached zenith after being cultured for seven days. But that of OP- 1/hydrogel group reached zenith after being cultured for ten days. The expression of ALP and Cbα1 were not distinctly different between OP-1 group with OP-1/hydrogel group after being cultured for fourteen days (P〉0.05). These results revealed that the polypeptide hydrogel delivery system combined with OP-1 can release OP-1 slowly to promote the osteoblasts to differentiate and increase the expression of ALP, OC and Cbfα1 of them.
