摘要
对三孢布拉霉菌发酵产β-胡萝卜素的最优条件进行研究,比较不同种子培养基、碳源、氮源、植物油、装液量、培养时间等对菌体生物量和色素形成的影响,通过正交试验确定最佳摇瓶发酵条件组合。结果表明,三孢布拉霉菌的最佳摇瓶发酵条件为:玉米淀粉2.4%、棉籽油4%、黄豆粉2%、柠檬酸钠0.3%、Tween200.15%、磷酸二氢钾0.2%、硫酸镁0.2%、VB10.0025%,以1∶4的正、负菌比例和20%的接种量接种,并在28℃、170r/min的条件下培养6d所得的生物量和色素产量分别为2.03g/50mL和1.216g/L。使β-胡萝卜素的生物量和色素产量分别提高了16.67%和110%。
The purpose of the paper is to study the optimum fermentation conditions on β-carotene production of Blakeslea trispora. The effects of carbon sources, nitrogen sources, vegetable oil, medium capacity and culture time on cell biomass and carotenoid production were investigated, and the optimal combination was received through orthogonal test. The result showed that the optimal fermentation conditions for β-carotene production were as follows: Cornstarch 3%, soybean powder 2%, cottonseed oil 3%, sodium citrate 0.3%, H2KPO4 0.2%, MgSO4 0.2%, VB1 0.0025%, Tween20 0.15%. medium volume 50 mL/250 mL, temperature of fermentation is 28 ℃, RPM is 170 r/min and culture time 72 h. The cell biomass, IS-carotene yield could reach 2.03 g/50 mL, 1.216 g/L respectively under the selected optimum condition, increase cell biomass and β- carotene yield by 16.67% and 110% respectively.
