摘要
研究热水浸提杏鲍菇中粗多糖的提取工艺路线,多糖含量用苯酚-硫酸法测得。在单因素试验的基础上,选定料液比、提取时间及醇沉浓度3个因素的3个水平进行中心组合试验,建立了多糖含量的二次回归方程,进一步优化了提取工艺条件,验证了模型的有效性,通过响应面分析以及岭嵴分析得到了优化组合条件。试验结果表明,醇沉浓度、提取时间对多糖得率都有显著影响,而料液比没有显著影响。当提取工艺条件为提取时间2.5h,料液比1∶38和醇沉浓度为91%时,水提杏鲍菇多糖得率达到极大值。此条件下多糖得率预测值为6.58%,验证值为6.52%。
The optimal craft process of extracting polysaccharides from Pleurotus eryngii by hot water was studied in this paper. The content of polysaccharides was determined by colourimetry using sulfuricacid and phenylicacid. Then the optimal conditions were obtained by response surface methodology on the basis of single factor test. The adequacy of the model equation for predicting the optimum response values was verified effectively by the validation data. By analyzing the response surface plots and the quadratic regression equations, the results indicate that the optimal conditions is following: time 2.5 hours, the volume of ethanolis 91% compared to the volume of extracting solvent and the ratio of water to material is 1:38. In this condition, the predicted value of the maximal yield of polysacchaddes is 6.58% while the effective value is 6.52%.
