摘要
目的观察乌天麻(Grastrodia elata bl)对大鼠心肌缺血再灌注I/R损伤的影响。方法灌胃给药14 d后,进行Langendorff离体心脏灌流,平衡、全心缺血(停灌)和再灌注各30 min,观察药物对左室发展压(LVDP)、冠状动脉流量(CF)、心肌组织超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量以及超微结构的影响。结果乌天麻明显抑制I/R引起的LVDP和CF的下降;提高心肌SOD活力和降低MDA含量,与模型组相比差异有极显著性意义(P<0.01),但没有明显的量效关系;明显减轻I/R引起的心肌超微结构的损伤性改变。结论乌天麻对I/R引起的心肌损伤有明显的拮抗作用,其机制可能与其抑制自由基生成,改善能量代谢等有关。
Objective To characterize the effect of Grastrodia Elata Bl on the ischemia/reperfusion I/R injury in rat hearts. Methods The isolated hearts were perfumed in the Langendorff mode and were subjected to either continuous perfusion for 90 rain or 30 rain for balance, global ischemia and reperfusion respectively. I.eft ventricular developed pressure (LVDP), coronary flow(CF), SOD activity and MDA content in myocardial homogenate, and the myocardial uhrastructure was also observed. Results It was found that Grastrodia elata B1 could significantly antagonize the decrease of LVDP and CF induced by I/R. Malignant arrhythmia incidence in Grastrodia Elata Bl group was lower than in I/R group with the difference being significant between them. Grastrodia Elata Bl could protect endogenous antioxidant enzymes, such as superoxide dismutase, and inhibit biomembrane lipid peroxidation, but there was no dove dependent relationship between these two drag groups. It was also found that myocardial ultrastructural changes induced by I/R were prevented in Grastrodia Elata Bl group. Conclusion Grastrodia clara bl could protect myocardium against I/R injury in rats, which might be related with its effects of restoring cardiac function.
