摘要
[目的]研究牡丹离体培养技术,解决牡丹快速繁殖问题。[方法]以MS为基本培养基,添加不同浓度的NAA、BA及IAA,对牡丹的胚进行体外培养。[结果]在没有添加任何激素的培养基上,离体胚生长类似种子萌发时胚的生长模式,子叶扩大伸长,达到约种子大小时停顿,根伸长生长极其显著;BA促进子叶膨大生长,当BA浓度增大时根生长受到抑制;NAA促进愈伤组织形成,抑制根生长。0.1mg/LBA促进牡丹胚的胚轴、根生长,有利于幼苗形成。[结论]为牡丹扩大繁殖提供了可行性途径。
[ Objective ] The research aimed to study the techniques of isolated culture of peony to implement rapid propagation. [Method ] Take MS as the basic culture medium,in vitro culture of peony' s embryo was carried on by adding different concentration of NAA, BA and IAA. [ Result ] Grow of vitro embryo was similar to the grow pattern of embryo when it was germinating on the culture medium which has no hormone, cotyledon expanded and grown, arid stopped growing when it achieved approximately seed size; cotyledon growth was improved by BA, the growth of root was inhibitted when consdntration of BA increased; NAA could improve the fromation of callus and inhibit the growth of root. Growth of hypocotyl and root of peony embryo were improved by 0.1 mg/L BA, which was conducive to the fromation of seedlings. [ Conclusion ] It was provided the feasible way for expanding propagation of peony.
