百喜草耐盐性愈伤组织的生理生化特性研究

摘要 [目的]从细胞水平分析比较百喜草耐盐性愈伤组织的生理生化特性。[方法]应用不同浓度(0、5、10、15、20g/L)的NaCl处理百喜草成熟种子,诱导产生耐盐的颗粒状愈伤组织和同种普通愈伤组织,研究其含水量、细胞膜透性、MDA含量、POD酶活性、SOD酶活性的变化。[结果]结果表明:随NaCl胁迫浓度的升高,百喜草耐盐组愈伤组织与对照组愈伤组织在多方面存在明显差异,前者含水量、丙二醛含量均高于后者;POD酶活性、SOD酶活性亦均高于对照组。在提高愈伤组织耐盐性诸因素中,脯氨酸起着重要作用。[结论]该研究为进一步筛选百喜草耐盐突变细胞系奠定了良好的基础。 [ Objective ] The research aimed to compare the physiological and biochemical characteristics of salt tolerance bahiagrass callus from the cell level. [ Method] Mature seeds of bahiagrass were dealed with different concentrations (0,5,10,15,20 g/L) of NaCl to induce salt tolerance granular callus and the kinds of general callus. And then the changes of water content, membrane permeability, MDA content, POD activity, SOD activity were studied. [ Result ] The results showed that, with the increase of NaCl concentration, there was an obvious difference in many ways between the salt group bahiagrass callus and callus of the control group. And the water content, MDA content of the former was obviously higher than the latter. The POD activity, SOD activity of the former was also higher than the latter. The proline played an important role in increasing salt-tolerant of bahiagrass callus. [ Conclusion] This study laid a good foundation for further screening of salt-tolerant mutant bahiagrass cell line .
