摘要
[目的]为深入研究菘蓝对根腐病的抗性机制提供理论依据。[方法]采用不同浓度菘蓝根腐病菌毒素处理菘蓝幼苗,筛选最适处理浓度;对最适浓度毒素处理过的菘蓝幼苗的外部形态进行鉴定,并测定其可溶性蛋白的含量。[结果]稀释10、20倍毒素处理过的菘蓝幼苗在试验期间全部萎蔫死亡,稀释40、50倍毒素处理过的菘蓝幼苗在试验期间大部分无病变现象,稀释30倍毒素处理过的菘蓝幼苗病变现象明显,处理72h后50%幼苗萎蔫死亡。经稀释30倍毒素处理的菘蓝幼苗可溶性蛋白的谱带大体相同,但也出现了个别特异性条带;经毒素处理的样品蛋白条带数多于对照,且染色较深。[结论]菘蓝根腐病菌毒素的筛选压力以稀释30倍最好。
[ Objective ] The aim was to provide the theoretical basis for further study on resistance mechanism of lsaris indigotica Fort. on root rot disease. [ Method] L indigotica seedlings were treated by pathotoxin produced by root rot fungus in L indigotica with different eonen. , and the optimal treatment concn, was selected. The external morphologies of L indigotica seedlings treated by pathotoxin with the optimal eonen. were identified, and the soluble protein contents in L indigotica seedlings were determined. [ Result] The L indigotica seedlings treated by pathotoxin which was diluted 10 and 20 times were all wilting and dead during test periods, and those treated by pathotoxin which was diluted 40 and 50 times had no lesions phenomenon during test periods and those treated by pathotoxin which was diluted 30 times had obvious lesions phenomenon, and 50% seedlings were wilting and dead after 72 h of the treatment. The protein bands of L indigotica seedlings treated by pathotoxin which was diluted 30 times were roughly equal, but there were some specific bands appeared also. Protein bands of the samples treated by pathotoxin was more than that of CK, and the stain of the bands was relatively dark. [ Conclusion] Diluting 30 times was the best screening stress of pathotoxin produced by root rot fungus in L indigotica.
