摘要
目的探讨内皮祖细胞(EPC)移植治疗糖尿病兔模型下肢缺血的疗效。方法将日本大耳白兔随机分3组:糖尿病PBS对照组(A组)、糖尿病内皮祖细胞移植治疗组(B组)、正常血糖内皮祖细胞移植治疗组(C组)。骨髓来源的内皮祖细胞经体外扩增培养7d后,通过肌内注射进行细胞移植。结果EPC移植后14d,彩超示B组兔缺血侧/正常侧下肢胫前动脉收缩期峰值流速比值明显增加,动脉造影示该组缺血侧下肢动脉显影血管数多于对照组(P<0.05)。结论 EPC移植能有效治疗糖尿病兔模型下肢缺血。
Objective To explore the effects of transplantation of endothelial progenitor cells .from autologous bone marrow on hindlimb ischemia in diabetic rabbit model. Methods Rabbits were randomly allocated into three groups:PBS control group (n= 6, A group), EPC transplantation in alloxan-induced diabetes rabbits (n= 8, B group), EPC transplantation in normal rabbits (n= 8, C group). Diabetic rabbit model was established by injecting alloxan, and hindlimb ischemia was induced by complete excision of femoral artery. Rabbits with hindlimb ischemia were treated by transplantation of ex vivo expanded EPC, and then effects were assessed by ultrasonography and angiography. Results Blood flow in ultrasonography was expressed as a ratio of ischemic to nonischemic leg in the same rabbit. The ratio was significantly increased in EPC transplantation rabbits with versus without diabetes at 14th day(0. 49±0. 05, 0. 57±0. 08,0. 60±0. 09). Angiography showed that angiogenesis index was significantly higher in EPC transplantation rabbits with diabetes versus control at 14th day. (6.25 ± 2.82, 10.25 ± 3.28, 10.13 ± 2.36). Conclusions Endothelial progenitor cells transplantation is an effective therapeutic method for treatment of hindlimb ischemia in diabetic rabbit model, and blood flow and angiogenesis index are increased significantly.
