摘要
目的探讨阿托伐他汀(ATO)和罗格列酮(RGZ)及二者联合对2型糖尿病大鼠心肌胶原堆积的影响。方法成年SD大鼠,通过高糖、高脂饮食加腹腔注射小剂量STZ制成2型糖尿病大鼠模型,并以ATO(20mg·kg-1·d-1)、RGZ(2mg·kg·d-1)及两药联合进行干预。光镜、电镜观察大鼠心脏的形态学改变及炎症细胞浸润程度,Masson染色观察心肌胶原沉积情况,生化法检测心肌羟脯氨酸含量。结果 RGZ及ATO均可在相似程度上减轻间质胶原沉积,两药联合较任一单药作用更明显(P<0.01)。结论 ATO及RGZ可减轻糖尿病大鼠心肌组织炎症细胞浸润和胶原沉积。二者联合可提高抗炎、抗纤维化程度。
Objective To study whether rosiglitazone, atorvastatin, and their combination can affect collagen accumulation in rats with type 2 diabetes. Methods The experimental type 2 diabetic rats were yielded by injecting streptozotocin(STZ, 30mg/kg) and feeding with high fat and glucose food, then treated by atorvastatin(20mg · kg^-1· d^-1 ), rosiglitazone(2mg · kg^-1· d^-1), and their combination. The morphology and inflammatory ceils infiltration in cardiac tissue were observed by light and electron microscope, and mycoardial collagen accumulation by Masson staining. The hydroxyproline content of cardiac tissue was examined by biochemical technique. Results The interstitial collagen accumulation and thickened capillary basement membrane were obviously relieved in the atorvastatin(Masson stain: 0.80±0. 16), rosiglitazone (0.78±0. 04) group and the combined two drug group(0.42±0. 06) compared with DM (1.36 ± 0. 16) group (all P 〈 0.01). Conclusions Atorvastatin and rosiglitazone may obviously relieve the collagen accumulation and inflammatory cells infiltration, thus delay the progress of the diabetic cardiomyopathy. Atorvastatin in combination with rosiglitazone may enhance effects of anti- inflammation and anti-fibrosis.
