PRL-3在乳腺癌癌变过程中的表达及意义

摘要 目的研究PRL-3在乳腺癌癌变过程中的表达及其与临床病理特征之间的关系。方法采用免疫组织化学方法检测60例乳腺癌组织及western blot方法检测27例乳腺癌及相应癌旁组织中PRL-3蛋白的表达,应用χ2及t检验分析其在乳腺癌组织中表达的意义及与临床病理特征之间的关系。结果免疫组化结果显示PRL-3蛋白在乳腺癌癌变过程中表达阳性率呈递增趋势;乳腺浸润性导管癌中PRL-3与患者的年龄、肿瘤大小、有无家族史、临床分期无明显关系,但与有无淋巴结转移、组织学分级相关。Western blot结果显示癌组织中PRL-3的表达明显高于癌旁乳腺组织,且有淋巴结转移组高于无淋巴结转移组。结论PRL-3蛋白的表达水平在乳腺癌癌变过程中呈现上升趋势,在一定程度上提示乳腺癌的发生发展及转移潜能。 Objective To study the expression of PRL-3 and the interaction with clinical-pathological features in breast cancer progression. Methods The expression of PRL-3 was examined in 60 paraffin-embedded specimens and in breast cancer progression and 27 freshly-taken tissues by Western Blot. The relationship between PRL-3 expression and clinic-pathological factors was analyzed by Chi-square and t test. Results The positive rate of the expression of PRL-3 protein showed a gradual increase in breast cancer progression. The PRL-3 expression had no relationship with age, gender, family disease history, tumor size, clinical stage, etc. However, the protein had a correlation with histological grade and lymph node metastasis. Western Blotting analysis of the protein level in the breast invasive and adjacent breast tissues showed there was a significant increase in the prolein exaression in breast cancer tissue compared with the adjacent breast tissue. Conclusion The results strongly suggest that the level of the PRI,-3 protein can indicate the potential of breast cancer to some extent and it may accelerate growth and metastasis of breast carcinoma.
