摘要
目的探讨天花粉蛋白(TCS)对人宫颈癌Caski细胞生长的抑制作用及量-效关系。方法不同浓度TCS作用于体外培养人宫颈癌Caski细胞株,采用吖啶橙荧光染色,荧光显微镜观察TCS对Caski细胞的生长抑制作用;透射电镜和TUNEL检测法观察Caski细胞凋亡及TCS诱导Caski细胞凋亡的量效关系。结果TCS对Caski细胞的生长具有明显的抑制作用,透射电镜和TUNEL法证实该抑制作用是通过诱导细胞凋亡实现的。在一定浓度范围内,细胞凋亡数与TCS的浓度之间有剂量依赖性。结论TCS能诱导体外培养Caski细胞凋亡,最佳浓度为60μg/ml。
Objective To study the inhibitory effect of different dose trichosanthin(TCS) on Caski cells.Methods The antiproliferative effect of different dose TCS on Caski cells was observed by fluorescence microscope.The TUNEL assay and electromicroscope were used for measurement of apoptosis of different dose TCS on The Caski.Results The apoptosis of different dose TCS on Caski Cells acted as the antiproliferation.The quantity of the apoptosis of the Caski cells depended on TCS dose within certain range. Conclusion The apoptosis of the Caski cells will happen when they were dealt with TCS and the best dose of the TCS is 60 μg/ml.
