摘要
目的研究大鼠口服清脑宣窍方有效部位后进入脑组织的清脑宣窍方移行成分。方法大鼠脑样品取出,洗去残血、匀浆、甲醇提取、离心后水溶,用C18固相萃取柱处理,选用色谱柱:Agilent Eclipse XDB-C18(4.6 mm×250 mm,5μm),Eclipse XDB-C18保护柱。建立大鼠口服清脑宣窍方有效部位后脑组织的LC-MS指纹图谱,并结合前期已分得的单体,对脑组织中移行成分进行了指认。结果确定了清脑宣窍方有效部位的靶器官脑的18个移行成分。结论清脑宣窍方栀子与三七的指标性有效成分栀子苷、人参皂苷Rg1、人参皂苷Rb1均能进入脑,发挥作用。
Objective To research on the new constituents in brain after rats taking effective fraction of Qingnaoxuanqiao formula orally.Methods The rat brain samples were taken out,cleaned remaining blood,smashed into plasm,extracted by MeOH,and dissolved by water after centrifuging.A column of C18 solid phase was used for the extraction.The procedure was carried out on a chromatography column of Agilent Eclipse XDB-C18 chromatography column(4.6 mm×250 mm,5 μm) and a protective column of Eclipse XDB - C18. Established LC - MS fingerprint of brain of rat after rats taking effective fraction of Qingnaoxuanqiao formula orally, and confirmed the constituents, using the monomer got previously. Results Confirmed 18 main fingerprint peaks in target tissue: brain. Conclusion The indicative effective constituents of Qingnaoxuanqiao effective fraction: geniposide, ginsenoside Rg1, ginsenoside Rbl and notoginsenoside R1 can be found in the target tissue: brain and exert pharmacology effection.
