靶向CXCR4的siRNA对人食管鳞癌EC9706细胞裸鼠移植瘤生长的抑制作用 被引量:3

摘要 目的探讨CXCR4siRNA对人食管鳞癌裸鼠移植瘤生长的抑制作用。方法建立人食管鳞癌细胞株EC9706裸鼠移植瘤动物模型,瘤旁注射化学合成CXCR4siRNA,同时以瘤旁注射阴性对照siRNA和PBS为对照。监测肿瘤生长变化,测量瘤质量。HE染色观察肿瘤病理学变化,RT-PCR、免疫组化方法检测CXCR4mRNA和蛋白变化。结果裸鼠体内实验结果显示,与对照组比较,CXCR4siRNA组肿瘤生长受到明显抑制,瘤质量下降;HE染色显示对照组肿瘤体积大、异形明显;RT-PCR、免疫组化结果表明CXCR4siRNA处理组CXCR4mRNA和蛋白表达下调。结论CXCR4siRNA能抑制人食管鳞癌EC9706细胞株裸鼠皮下移植瘤的形成,且能在体内有效下调CXCR4mRNA和蛋白的表达。 Objective To investigate the inhibitory effect of CXC chemokine receptor 4 ( CXCR4) siRNA on the growth of transplanted tumors of esophageal squamous carcinoma in nude mice. Methods Nude mice model bearing esophageal squamous carcinoma was reproduced by implanting human esophageal squamous carcinoma cell line EC-9706. CXCR4 siRNA was injected to the area nearby the tumors. The tumors injected with control siRNA or PBS served as control. Tumor growth and tumor weight were measured. Histopathological changes of the tumors were observed with HE staining,and the mRNA and protein expressions of CXCR4 were detected by RT-PCR and immunohistochemical staining respectively. Results Tumor growth was obviously suppressed and tumor volume was significantly smaller in the group treated by with CXCR4 siRNA than in control groups. While the histopathological features in the control group showed that the tumor volumes were bigger and large amount of atypic tumor cells was seen. RT-PCR and immunohistochemical staining indicted that CXCR4 mRNA and protein expressions were reduced in in the group treated by with CXCR4 siRNA. Conclusion CXCR4 siRNA inhibits the tumorigenesis of EC9706 cell xenograft in nude mice and downregulates the CXCR4 mRNA and protein expressions in vivo.
