摘要
目的观察血管内皮生长因子(vascular endothelial growth factor,VEGF)在前列腺增生(BPH)及前列腺肿瘤中的表达,探讨其在前列腺疾病发生、发展中的作用。方法以临床收集到的确诊为前列腺增生37例、前列腺癌12例病变组织及7例来自意外死亡成年男性志愿者的正常前列腺组织共56例标本作为研究对象。以HE染色观察不同病理状况下前列腺的组织结构改变;以免疫组织化学染色法定位并定量VEGF在前列腺组织中的表达,并比较各组间表达的差异性。结果 HE染色显示,前列腺增生及前列腺癌病变表现为特征性的组织病理学改变。通过免疫组织化学染色观察到VEGF弥漫分布于前列腺上皮细胞的胞浆、基质细胞和间质,在微血管内皮细胞及其周围基质细胞中呈强阳性表达;VEGF在3种前列腺组织中均有表达,但前列腺增生与前列腺癌组表达高于正常对照组,经秩和检验差异具有显著性(P<0.05)。结论正常前列腺组织有VEGF的表达,表明其在维持正常机体前列腺的完整性方面发挥一定的作用。前列腺增生及前列腺癌组织VEGF表达较正常前列腺组织明显增多,提示血管形成促进因子在这两种疾病的发生、发展过程中可能起一定的促进作用。
Objective To observe the expression of vascular endothelial growth factor ( VEGF ) in benign prostate hyperplasia ( BPH ) and prostate cancer, so as to investigate its possible function in prostate diseases. Methods The tissue proved by pathology from thirty seven patients with BPH and twelve with prostate cancer were collected from clinic. The normal prostate tissue from seven male donors who died unexpectedly were collected. The hematoxylin eosin method was used to observe the histopathological changes,immunohistochemical staining of VEGF in prostate tissue was taken. Results The specimens of BPH and prostate cancer presented distinctive histopathological changes. VEGF were widespread on endochylema of prostate epithelial cells, stroma cells and matrix. The high expression level was also found in capillary vessel endothelial cells and surrounding stroma cells. VEGF was expressed among the three groups, however, there was higher expression level in the BPH and prostate cancer group than that of the control group, which had statistical differences ( P 〈 0.05 ). Conclusions The expression of VEGF in normal prostate tissue may play physiological role in maintaining the integrity and functions of prostate. The higher expression level of VEGF in BPH and prostate cancer tissue may have inducible roles in disease aggravation.
