摘要
目的探讨桂皮醛溶液用于大鼠牙周炎局部药物治疗的效果。方法通过丝线结扎双侧上颌第二磨牙和喂饲高糖水建立大鼠实验性牙周炎动物模型。实验动物随机分成1个对照组和3个实验组(T1、T2、T3组),每组5只。将生理盐水和32、64、128mg/L的桂皮醛溶液分别作为对照组和T1、T2、T3组的牙周冲洗用药,在第1、4、7天对各组大鼠行牙周炎的局部药物治疗。用滤纸条取治疗前(基线期)和治疗后第14天大鼠患牙龈沟液,检测龈沟液量并用酶联免疫吸附法检测白细胞介素(interleukin,IL)-1β浓度。结果牙周局部药物治疗前,4组龈沟液量的差异无统计学意义(F=2.766,P>0.05),4组龈沟液IL-1β水平的差异(F=0.250,P>0.05)无统计学意义。治疗后14d,对照组、T1、T2、T3组龈沟液量分别为(0.496±0.061)、(0.417±0.079)、(0.428±0.109)、(0.308±0.047)μL;龈沟液中IL-1β水平分别为(20.894±5.202)、(20.468±3.199)、(17.770±4.024)、(14.432±5.955)pg/L。同治疗前相比,治疗后各组龈沟液量减少和龈沟液中IL-1β水平降低均有统计学意义,其中T3组降低最为明显(t=18.939,P=0.000;t=6.661,P=0.000)。治疗前后龈沟液量差值在各实验组和对照组之间的差异均有统计学意义;各实验组间两两比较,治疗前后龈沟液量差值在T1组和T2组之间的差异无统计学意义,在T3组和T1组(P=0.001)、T3组和T2组(P=0.015)之间的差异有统计学意义。治疗前后IL-1β水平差值在各组间两两比较的差异均无统计学意义,但差值在T3组与对照组之间的差异最大(P=0.070)。结论浓度为32、64、128mg/L的桂皮醛溶液对大鼠实验性牙周炎均有缓解作用;128mg/L是否为最佳适用浓度尚有待进一步研究证实。
Objective To study the effects of cinnamicaldehyde solution in the use of local medication of periodontitis on rats. Methods To build an animal model of rat experimental periodontitis by inducing its maxillary second molars by silk-ligature and feeding high carbonhydrate water. One control group and three experimental groups ( Group T1, T2, T3) , five in each group, were parted at random, irrigated the tooth periodontal pockets by using normal saline and 32, 64, 128 mg/L cinnamicatdehyde solution on the first, fourth, seventh day respectively. Take the rat gingival cervieular fluid(GCF) of the pre-therapy (on baseline) and the fourteen-day after therapy by using periopaper to detect the volume of GCF and interleukin-1β (IL-1β) concentration through ELISA. Results Before the medication, there is no statistical significance in volume( F = 2. 766 ,P 〉 0.05 ) and IL-1β level ( F = 0. 250, P 〉 0.05 ) of the four GCF, but after 14-day period, the volumes of GCF in control group, group T1, T2, T3 are (0. 496 ±0.061 ), (0.417 ±0. 079), (0.428 ±0. 109), (0. 308 ±0. 047 ) μL; the IL-1β levels are ( 20. 894 ±5. 202 ), ( 20. 468 ± 3. 199 ), ( 17. 770 ±4. 024 ), ( 14. 432 ±5. 955 ) pg/L. Compared to the pre-therapy, the reducing of the volumes of GCF and the IL-1β levels in each group has statistical significance, among which the data of group T3 is distinct (t = 18. 939,P =0.000;t =6. 661 ,P = 0. 000). Statistical significance exists in the difference of volumes of GCF before and after the medication, among which group T3 and group Tt (P = 0. 001 ), group T3 and group T2 (P =0. 015 ), but not in the group T1 and group T2. Statistical significance does not exist in the difference of IL-1β levels before and after the medication, but the difference got biggest between group T3 and the control (P = 0. 070). Conclusion The 32, 64, 128 mg/L cinnamicaldehyde solution has corrective effect on rat periodontitis, but whether the 128 mg/L is the most suitable one or not needs further study.
