摘要
目的探讨慢性粒细胞白血病(CML)患者骨髓单个核细胞(BMMNC)中富半胱氨酸61基因(Cyr61)的表达水平及其临床意义。方法应用半定量RT-PCR和免疫组化技术分别检测38例CML患者和10例正常对照者(正常对照组)骨髓单个核细胞中Cyr61mRNA和蛋白水平的表达情况,CML患者中慢性期20例(慢性期组),加速期11例(加速期组),急变期7例(急变期组)。结果38例CML患者中,慢性期组、加速期组、急变期组Cyr61mRNA的表达明显高于正常对照组(P<0.05)。Cyr61基因在加速期及急变期的表达高于慢性期(P<0.05),Cyr61基因在加速期和急变期的表达虽升高,但两者差异无统计学意义(P>0.05)。Cyr61蛋白在CML中的表达明显高于正常对照组,CML加速期和急变期中Cyr61蛋白的表达高于慢性期(P均<0.05)。结论Cyr61基因参与了CML的发生、发展过程,且与CML的急变有关,可作为预示CML急变的实验依据。
Objective To investigate the chnical significance of cysteine rich 61(Cyr61)in the bone man'ow (BM) of chronic myelogenous leukemia(CML) patients. Methods Expressions of Cyt61 mRNA and Cytrl protein were detected in bone marrow mononuclear cells(BMMNC) from 38 patients with CML (20 chronic phase, 11 accelerated phase and 7 blast crisis) and 10 norreal controls by the reverse transcriptase -polymerase chain reaction (RT-PCR) method and immtmohistochemistry, respectively. Results Expression of Cyr61mRNA in a chronic phase, accelerated phase and blast crisis was significantly higher in CML patients than in normal controls( P 〈 0.05). Expression of the Cyr61 gene was higher in an accelerated phase and blast crisis than in the chronic phase( P 〈 0. 05), but showed no significance ( P 〉 0.05). Expression of Cyr61 protein was higher in CML patients than in normal controls(P 〈 0. 05). Expression of Cyr61 protein was higher in the accelerated phase and blast crisis than in the chronic phase( P 〈 0.05). Conclusion Over-expression of the Cyrrl gene which plays an important role in the acute transformation of chronic myelogenous leukemia may relate to the progression and genesis of CML and provides an experimental basis for early diagnosis of CML blastic crisis.
