摘要
目的:研究温阳化瘀中药复方对糖尿病大鼠脂肪组织抵抗素mRNA表达的影响,探讨温阳化瘀中药治疗糖尿病胰岛素抵抗的作用机制。方法:采用逆转录多聚酶链式反应(RT-PCR)方法测定糖尿病大鼠腹部脂肪组织和下肢骨骼肌间的脂肪组织抵抗素mRNA表达。结果:肥胖的2型糖尿病胰岛素抵抗模型大鼠具有脂肪源性抵抗素mRNA高表达的特点,而腹腔内脂肪组织的表达水平比周围组织骨骼肌肌间脂肪组织的表达水平更高。(P<0.01)温阳化瘀中药复方治疗可以降低脂肪源性抵抗素的mRNA的表达水平。结论:温阳化瘀中药通过降低脂肪源性抵抗素的mRNA的表达水平治疗胰岛素抵抗。
Objective:Research the effect of Warming-yang Dissolving-stasis CCTM on resistin mRNA expression in adipose tissue of diabetic rats, to investigate the mechanism of Warming-yang Dissolving - stasis CCTM improving insulin resistance. Methods :The reverse transcription-polymerase chain reaction( RT -PCR) was adopted to detect the expressions of resistin gene in abdomen and leg intermuscular fatty tissues. Resu/ts : Fat-derived high expression of resistin mRNA was character in obese type 2 diabetes mellitus rats. Resistin mRNA expression was higher in abdomen than leg intermuscular fatty tissues (P 〈 0. 01 ). Treatment by Warming-yang Dissolving-stasis CCTM could decrease fat-derived resistin mRNA expression. Conclusion:Warming-yang Dissolving-stasis CCTM improve insulin resistant by decreasing fat-derived resistin mRNA expression.
