摘要 [目的]研究多花胡枝子(Lespedeza floribunda)组培再生体系。[方法]以子叶节为初代分化外植体,诱导不定芽,通过继代培养获得再生茎段,培养生根后获得再生植株。[结果]最佳种子消毒时间为0.1%HgCl2溶液消毒8min;初代分化选择子叶节为外植体时,最佳分化配方是1/2MS+6-BA1.0mg/L+NAA0.05mg/L+IBA0.2mg/L;最佳茎段继代配方是WS+6-BA0.7mg/L+NAA0.1mg/L+IBA0.1mg/L;最佳生根配方是WS+IBA0.3mg/L,生根率为74.8%。6-苄基腺嘌呤(6-BA)对初代分化与继代分化的影响均达显著水平(α=0.05)。与萘乙酸(NAA)相比,吲哚丁酸(IBA)诱导多花胡枝子生根的能力更好。 [ Objective ] The aim was to study regeneration system of Lepedeza floribunda. [ Method ] Cotyledonary node was adopted as primary differentiation explant to induce adventive buds. Regenerated plants were obtained after rooting culture of shoots which acquired in sec- ondary differentiation. [ Result] The optimum time of seed disinfection was 0.1% HgCl2 solution disinfected 8 min. Primary differentiation acquired cotyledonary nodes as explant and the optimal prescription was half strength MS medium plus 6-BA 1.0 mg/L, NAA 0.05 mg/L,and IBA O. 2 mg/L. The suitable medim of secondary differentiation for propagated shoots was WS medium containing 6-BA O. 7 mg/L + NAA 0.1 mg/L + IBA 0.2 mg/L. The efficient rooting prescription was WS medium with IBA O. 3 mg/L and rooting ratio was 74.8%. The effect of 6- benzylaminopurine (6-BA) in primary differentiation reached the significant level (α = 0. 05 ) even secondary differentiation. Indole-3-butyric acid (IBA), better than Napthaleneacetic acid (NAA), was able to induce regenerated shoot rooting.
