摘要 [目的]找出一种适合杏鲍菇自身的分子标记方法,为其亲缘关系研究提供借鉴。[方法]利用随机扩增多样性DNA(RAPD)对23种来自我国不同地区的杏鲍菇品种进行遗传多样性分析,寻找一种适合杏鲍菇自身的分子标记方法。[结果]筛选出的12个RAPD引物扩增出谱带121条,多态性谱带95条,多态率为78%,表现出丰富的RAPD多态性;利用NTYS-PC2.1软件进行UPGMA聚类分析,可将该23份材料分为5大类,存在较大的遗传差异性。[结论]RAPD技术用于杏鲍菇遗传多样性的分析是有效的,并且能检测到更高的品种间遗传差异。 [ Objective ] The reference for the analysis of the relation of Pleurotus eryngii was provided through the selection of the suitable method of molecular marker. [ Method ] The genetic diversity of 23 species of Pleurotus eryngii from the different regions of China was analyzed with RAPD( randomly amplified polymorphic DNA) marker. [ ResultsI The result showed that 121 bands were amplified with 12 polymorphlc primers including 95 polymorphic ones and their polymorphism percentage was 78%. 23 samples could be clustered into 5 groups based on the UPGMA method with NTYS-PC2 and there was more bigger genetic diversity among them. [ Conclusion] The technique of RAPD marker was efficiency on the analysis of the genetic diversity of Pleurotus eryngii.
