摘要 [目的]通过透射电镜来观察黑亚14号不同类型愈伤组织的超微结构。[方法]采用常规电镜样品制备和观察方法。[结果]透射电镜观察结果表明,初始愈伤组织细胞有非常明显的液泡,细胞核被其挤压位于细胞的边缘一角,但仍可观察到核仁;胚性愈伤组织细胞壁与初始愈伤组织细胞壁相比加厚,在靠近细胞膜的胞质区域里有较多的淀粉粒,且可观察到其内含有淀粉粒的叶绿体;非胚性愈伤组织细胞基本只能观察到液泡,几乎无其他细胞器;褐化愈伤组织细胞器崩解,基本没有细胞器。[结论]亚麻不同类型愈伤组织在细胞超微结构水平上有显著的差异,可以作为反映愈伤组织细胞分化能力的指标。 [Objective] The aim was to observe the ultrastructure of different callus in Heiya No. 14 by transmission electron microscopy. [ Methods ] Sample preparation and observation all adopted conventional transmission electron microscopy. [ Results ] It was showed by trans- mission electron microscope that the initial callus cells had large central vacuole, which squeezed its cytoplasm to a thin layer around the brim of cell. Nuclear distributed the comer of cells because of being squeezed by large vacuole, however, it also found the presence of nuclear. Compared embryogenic callus with initial callus, its cell wall became thick,and had dense cytoplasm which was full of the whole cell, big nucleus with obvious nucleolus. Many starch grains and chloroplasts including starch grains distributed on the cytoplasm area of cell membrane. Non-embryoenic callus only had vacuole, and nearly no other organelle. Browning callus cell collapsed and had not cell organelle. [ Conclusion] Different type of flax callus have significant difference in the level of cell ultrastructure, which were used as index of callus cell differentiation capability.
