Skp2-siRNA干预对Eca-109食管癌细胞系p21和p27蛋白及细胞周期的影响 被引量:4

摘要 目的探讨Skp2-siRNA干扰后食管癌Eca-109细胞株Skp2、p21和p27蛋白表达情况及其对细胞增殖调控的影响。方法经Skp2-siRNA干扰食管癌Eca-109细胞株24、48和72 h后,用免疫细胞化学法检测Skp2、p21和p27蛋白的表达情况,流式细胞仪检测细胞增殖周期的分布。结果食管癌Eca-109细胞株主要分布在G1期,Skp2蛋白表达下降(P<0.05或<0.01),p21和p27蛋白的表达增高(P<0.05或<0.01)。结论运用Skp2-siRNA技术可以有效降低对p21和p27蛋白的降解,促进细胞停滞在G1期,降低细胞增殖活性。
