硫糖铝对大鼠胃溃疡黏膜选择性黏附的体外评价 被引量:5

Selective adhesion of sucralfate to gastric ulcer in vitro

摘要 目的:在模拟胃生理条件下,考察硫糖铝对大鼠胃溃疡黏膜的选择性黏附作用。方法:电感耦合等离子体-原子发射光谱法(ICP—AES)、紫外-可见分光光度法(UV)分别测定硫糖铝中铝离子(Al^3+)和八硫蔗糖在正常胃黏膜及胃溃疡黏膜的组织残留量。结果:ICP—AES、UV法测定Al^3+和八硫蔗糖的精密度、准确度及回收率均可满足实验要求;人工胃液pH对Al^3+和八硫蔗糖在正常胃黏膜、胃溃疡黏膜的组织残留量有明显的影响;Al^3+的组织残留量为pH1~3〉〉pH4〉pH5(P〈0.05),正常胃黏膜与胃溃疡黏膜Al^3+的组织残留量在相同pH差异无显著性;pH对八硫蔗糖的组织残留量影响与Al^3+相似,但在pH≤3时,八硫蔗糖在胃溃疡黏膜上的组织残留量显著高于正常胃黏膜(P〈0.05)。结论:ICP-AES、UV法测定硫糖铝中Al^3+和八硫蔗糖的胃组织残留量是准确可行的;硫糖铝中Al^3+对胃溃疡黏膜无选择性黏附作用,八硫蔗糖在pH≤3时对胃渍疡黏膜具有选择黏附作用。 OBJECTIVE To investigate the selective adhesion of sucralfate for SD rats' gastric ulcer mucosa in a simulated physiological conditions. METHODS The residuals of Al^3+ and sucrose octasulfate on gastric normal mucous and ulcer mucosa were determined by inductively coupled plasma-atomic emission spectrometry (ICP-AES) and ultraviolet-visible spectrophotometry (UV). RESULTS It could meet the test requirements that the determination precision, accuracy and recovery of Al^3+ and sucrose octasulfate by ICP-AES and UV. Artificial gastric juice pH had a significant impact for the residues of Al^3+ and su crose octasulfate on the gastric normal mucosa and ulcer mucosa. The residuals of Al^3+ was pH 1-3〉〉pH 4〉pH 5(P〈 0. 05), but no significant difference between gastric normal mucosa and ulcer mucosa at the same pH. It was similar to Al^3+ that the residuals of sucrose octasulfate was influenced by pH, but the residues of sucrose octasulfate in the gastric ulcer mucosa were significantly higher than the residues in normal mucosa in pH≤ 3 (P〈0. 05). CONCLUSION It is feasible that the Al^3+ and sucrose octasulfate are determined by ICP-AES and UV. Sucrose octasulfate has a selective adhesion to gastric ulcer mucous in pH≤3, but Al^3+ has the role of non-selective adhesion.
