乌司他丁对急性肺损伤组织中水通道蛋白及Fas表达的研究 被引量:4

摘要 目的探讨乌司他丁(UTI)对急性肺损伤组织中水通道蛋白-1、4(AQP-1、4)及Fas的表达及临床意义。方法应用脂多糖(LPS)诱导大鼠急性肺损伤(ALI)模型,然后应用乌司他丁进行干预。实验分为3组:实验对照组、模型组、乌司他丁组。采用免疫组化染色,检测UTI对急性肺损伤大鼠肺组织中AQP-1、4及Fas的表达,利用HPIAS-2000图像分析系统测定水通道蛋白-1、4和Fas在以上各组中表达的平均光密度和平均阳性面积率。结果实验对照组肺组织高水平表达AQP-1、4,Fas呈低表达;模型组肺组织呈浅黄色染色,AQP-1、4表达较低,Fas呈高表达;乌司他丁组肺组织高水平表达AQP-1、4,Fas呈低表达。经q检验,实验对照组与模型组之间,AQP-1、4及Fas的平均光密度及阳性面积率差异有统计学意义(P<0.01?,实验对照组与乌司他丁组之间,AQP-1、4及Fas的平均光密度及阳性面积率差异无统计学意义(P>0.05)。结论乌司他丁能使急性肺损伤组织中AQP-1、4呈高表达,Fas呈低表达,乌司他丁对急性肺损伤组织有重要的保护作用。 Objective To approach the express and clinical significance of AQP and Fas in acute lung injury tissue by ulinastatin. Methods Applying LPS to lead rats ALI model and intervene by UTI. The experiment was divided into three group., experiment contrast group,model group and ulinastatin group (UTI). Immunohistochemical stainings were performed to detect the expression of AQP and Fas in those three groups. The average optical density'and the rate of positive area of expression of AQP and Fas were analyzed using image analysis Image HPIAS-2000 Analysis System. Results The average optical density and the rate of positive area of expression of AQP-1,4 in experiment contrast group and in ulinastatin group (UTI) were significantly higher than those in model group (P〈0.01). No significant difference was found in the average optical density and the rate of positive area between the expression of AQP-1,4 in experiment contrast group and those in ulinastatin group (P〉0.05). The average optical density and the rate of positive area of the expression of Fas in experiment contrast group and in ulinastatin group (UTI) were significantly lower than those in model group (P〈0.01). No significant difference was found in the average optical density and the rate of positive area of between the expression of AQP-1,4 in experiment contrast group and that in ulinastatin group (P〉0.05). Conclusion UTI can lead to AQP-1,4 to offer high level the express and to Fas show low level express in ALT tissue. UTI has important protect function to ALI tissue.
