摘要
目的:优选龙胆中龙胆碱的最佳提取工艺。方法:以龙胆碱含量为指标,以氨水浓度、乙醇浓度、醇氨比例、提取时间为考察因素,采用正交试验优选最佳提取工艺。结果:最佳提取工艺为龙胆药材加10倍量的溶剂(95%乙醇-20%氨水(3∶1)),80℃回流提取2次,每次1h。结论:该工艺稳定、合理、可行,适用于工业化生产。
OBJECTIVE: To optimize the extraction technology of gentianine from Radix et Rhizoma Gentianae. METHODS: The extraction technology was optimized by orthogonal experiment with the content of gentianine and the yield of extractum as indexes while with ammonia concentration, alcohol concentration, the ratio of alcohol to ammonia and the extracting time as factors. RESULTS: The optimal extraction technology was adding 10-fold volume of solvent [95% alcohol- 20% ammonia (3: 1)] to herbal gentian for reflux extraction of 2 times (1 hour/time) at 80 ℃ . CONCLUSION: This extraction technology is stable, reasonable and feasible, and it is applicable for experimental studies.
