摘要
目的:检测重组人CEA痘苗病毒(HumanCEARecombinationVacciniaVirus,rV-CEA)在动物体内应用的安全性与可行性,为肿瘤的基因治疗提供实验依据。方法:将重组人rV-CEA和痘苗病毒野生型及生理盐水分别接种纯种新西兰兔,观察组织结构并用免疫组化方法检测兔体内人CEA的表达。结果:接种rV-CEA组的第8天,兔血清CEA含量明显升高,第15天后在免血清中测出抗CEA抗体。光镜观察接种rV-CEA兔的心、肝、脾、肾、淋巴结和皮肤等组织,其结构均无异常变化,电镜下亦未见肾小球基底膜有免疫复合物的沉积。免疫组化检测结果:在兔组织器官中均未检测到CEA的表达。结论:rV-CEA在体内应用是安全可行的。
objective: Safety and possibility of Recombination Human CEA Vaccinia Virus (rV - CEA) were detected in rabbits to proveide experimental data for gene therapy in cancer. Methods: The rabbits were vaccinated with either rV - CEA, the wild-type vaccinia virus or only saline. Histology was observed and human CEA expression was detected immunlhistochemically. Results: On day 8, high level of serum CEA could be detected. On day 15, antibody against CEA were detected the rabbits with rV- CEA. There was no change in the liver, kidney, spleen, skin and cardiac muscle under LM, and nor immune - complex deposition on glomerular basement membrane under EM. The immunohistochemical results showed that no CEA expression in waccinated rabbits tissue. Conclusion: Vaccinataion of rV - CEA is safe and efficient in vivo.
