硫酸镁对小鼠全脑缺血再灌注损伤模型NO、NOS的影响 被引量:1

摘要 目的:探讨不同剂量硫酸镁(MgSO4)对小鼠全脑缺血再灌注损伤的治疗作用。方法:将小鼠随机分成对照组、再灌注组、MgSO490 mg/kg组、MgSO4180 mg/kg组、MgSO4360 mg/kg组;结扎双侧颈总动脉和颈部软组织加压的方法造成全脑缺血,缺血30 min,再灌注60 min。监测脑电图变化,观察病理学形态变化;检测脑组织NO含量和一氧化氮合酶(NOS)、诱导型NOS(iNOS)活性。结果:MgSO4各治疗组与再灌注组比较,脑组织中NO含量减少(P<0.01),NOS和iNOS活性降低(P<0.01),脑组织病理损害减轻。结论:硫酸镁对脑缺血再灌注损伤有治疗作用,可能与降低NOS、iNOS活性,减少NO含量有关。 Objective:To explore the effect of different doses of magnesium sulfate (MgSO4 ) on reperfusion injury of mice with general cerebral ischemia. Methods:The mice were randomly divided into control group, ischemia reperfusion(I/R) group, MgSO4 90 mg/kg group, MgSO4 180 mg/kg group and MgSQ 360 mg/kg group. General cerebral ischemia model was established by blocking the bilateral common carotid arteries and pressing the soft tissues in carotid. The mice were reperfused for 60 min after 30 rain ischemia. The change of EEG was monitored and the pathological change in morphology was observed. The content of nitric oxidel (NO) and the activity of nitric oxide synthase(NOS) and inducible NOS(iNOS) were also detected. Results: Compared with MgSO4 groups,the content of NO (P 〈 0.01 ), the activities of NOS and iNOS in the I/R group were decreased( P 〈 O. 01 ), and the pathologic damage in brain tissues relieved. Conclusions:The curative effect of MgSO4 on cerebral ischemia-reperfusion injury might be associated to the decrease of the NO contents and the lowering of the activities of NOS and iNOS.
