摘要
探讨蜂胶对高龄小鼠抗衰老机理。以自然衰老小鼠为模型,分别用20,33,80 mg/kg剂量的蜂胶给小鼠灌胃24 d,MTT法观察脾脏淋巴细胞增殖;乳酸脱氢酶法(LDH)观察脾脏NK细胞及腹腔MΦ细胞对YAC-1的杀伤活性;胸腺细胞增殖法观察脾脏淋巴细胞IL-2及腹腔MΦ细胞IL-1的活性水平;采用化学比色法观察小鼠脑组织丙二醛(MDA)的含量和超氧化物歧化酶(SOD)的活力。试验结果表明,不同剂量蜂胶均可以提高小鼠抗氧化及免疫功能,其中中剂量蜂胶(33 mg/kg)明显提高小鼠脾脏淋巴细胞增殖(P<0.01)和NK细胞、Mφ细胞杀靶活性(P<0.01);同时,IL-1和IL-2产生也明显增加(P<0.01);丙二醛(MDA)的含量降低(P<0.01);超氧化物歧化酶(SOD)的活力显著增加(P<0.01)。适量剂量蜂胶具有明显提高高龄小鼠抗氧化及机体免疫功能的作用。
In this paper the anti-aging mechanism of proplis on aging mice were investigated. Nature old mice were used as aging animal model. The model animal was medicated with 20 mg/kg,33mg/kg and 80mg/kg proplis for 24 days, then, the splenetic lymphocytes proliferation was analyzed by MTT colorimetry, the activity of NK and Mφ were examined with the method of LDH, the production of IL-1 by peritoneal macrophages and IL-2 by splenetic lymphocytes were assayed by thymus cells proliferation method, the contents of MDA and the activity of SOD were tested with the methods of colorimetry. The results indicated that the splenetic lymphocytes proliferated, the activity of NK and Mφ, the production of IL-1 and IL-2 and the activity of SOD increased, but the contents of MDA declined when the mice were treated with 33mg/kg proplis, proplis contents might have the effects of antioxidization and increasing immune function in aging mice.
