摘要
用脂质体法将Smad2反义寡核苷酸瞬时转染人肾小球系膜细胞,设立正常糖组、高糖组、脂质体组、甘露醇组、转染Smad2正义寡核苷酸组及反义寡核苷酸组,ELISA法检测各组纤连蛋白和层黏连蛋白的含量。结果显示高糖培养可使人。肾小球系膜细胞分泌纤连蛋白和层黏连蛋白增加,转染Smad2反义寡核苷酸可抑制纤连蛋白和层黏连蛋白的分泌。
Sense and antisense oligodeoxynucleotides (ODN)were synthesized and transferred transiently into human renal mesangial ceils through lipofectamine 2000. There were 6 groups incubated differently : with normal glucose, high glucose, lipofectamine 2000, mannitol, sense ODN, and antisense ODN, Fibronectin ( FN ) and lamimin (LN) were examined by ELISA. The result revealed that human renal mesangial cells incubated with high glucose can yield increased secretion of FN and LN, while Smad2 antisense ODN can restrain the secretion of FN and LN.
