摘要
目的运用基因芯片筛选与炎性衰老相关的炎性细胞因子与受体特征基因,探讨其炎性细胞因子网络调控机制;观察淫羊藿总黄酮(EF)和淫羊藿苷(Ica)对该网络的干预效果与机制。方法SD大鼠分为4月龄(4m)、24m、24m+EF和24m+Ica组,每组10只。分别取各组大鼠海马和肺组织,用炎症细胞因子与受体基因芯片对各组组织分别进行基因表达检测。结果(1)海马组织:24m组与4m组比较,促炎症细胞因子表达上调;同时抗炎症细胞因子下调。24m+EF组、24m+Ica组与24m组比较,可见大量抗炎症细胞因子上调而促炎症细胞因子下调。(2)肺组织:24m组与4m组比较,促炎症细胞因子上调,同时抗炎症细胞因子下调。24m+EF组、24m+Ica组与24m组比较,发现大量抗炎症细胞因子上调而促炎症细胞因子下调。结论老年大鼠存在促炎性细胞因子基因表达上调和抗炎性因子表达下调,导致促-抗炎性细胞因子网络平衡失调;EF和Ica可能通过下调促炎性细胞因子基因表达和上调抗炎性细胞因子基因表达,重塑促-抗炎性细胞因子网络平衡而干预炎性衰老。
Objective To study the mechanisms of inflammatory cytokines network regulation in the development of inflamm-aging rats and analyze the Epimedium total flavonoids(EF) and Icariin(Ica) intervention outcome.Methods Healthy male Sprague Dawley rats were divided into 4 months(4 m),24 m,EF+24 m(24 m rats treated with EF) and Ica+24 m groups(24 m rats treated with Ica),each with 10 rats.The genes expressions of hippocampus and lung tissues from the rats in each group were analyzed by inflammatory cytokines and receptors cDNA microarray respectively.Results(1)In rat hippocampus tissues,compared with those in 4 m group,pro-inflammarory genes were up-regulated and anti-inflammatory genes were down-regulated.After treated by EF or Ica,anti-inflammatory genes were up-regulated while pro-inflammarory gene were down-regulated in 24 m group.(2)In rat lung tissues,compared with those in 4 m group,24 m rats showed up-regulation of pro-inflaming genes,while anti-inflammatory genes were down-regulated.Similar results were observed after intervention by EF and Ica.Conclusions Up-regulation of pro-inflamatory cytokines or receptors together down-regulation of anti-inflamatory genes may eventually contribute to disequilibrium of the network of pro-and anti-inflammatory cytokines and receptors and high inflammatory state.EF and Ica may interfere in inflamm-aging through rebuilding the new equilibrium of the network of pro-and anti-inflammatory cytokines and receptors.
