大鼠急性缺血性肾功能衰竭血管内皮生长因子水平的变化

摘要 目的探讨大鼠肾脏缺血/再灌注损伤(IRI)后不同时段血管内皮生长因子(VEGF)浓度的变化在急性肾衰竭(ARF)中的发病机制。方法建立大鼠肾脏IRI模型,采用ELISA法检测IRI术后不同时间段血、尿VEGF水平,并与肾功能做相关分析。结果手术组术后6h血清尿素氮(BUN)、肌酐(SCr)、血钾显著升高,至术后48h达高峰,从术后72h开始回落(P<0.05)。手术组术后6h血清VEGF显著升高,术后12h达高峰,从术后24h开始下降至正常;手术组术后6h尿液VEGF显著升高(P<0.01),至术后24h降至低于假手术组,从术后48h开始回升。血VEGF水平与尿VEGF水平呈正相关,与血钾、BUN、SCr水平无显著相关(P>0.05)。结论血、尿VEGF升高水平可以作为ARF大鼠肾脏IRI早期肾脏缺血缺氧的监测指标。 Objective To investigate the serum and urine concentration of vascular endothelial growth factor(VEGF) in rat with ischemia/reperfusion injury.Methods Rat renal I/R model was performed by placing the atraumatic microvascular clamp in both renal artery for 60 minutes.VEGF in serum and urine were detected at various intervals with ELISA.Relationship between VEGF and renal function was measured.Results Serum BUN,Cr and kalium significantly increased from 6 h to 72 h post-reperfusion(P〈0.05),reached its peak on 48 h.Serum VEGF significantly increased from 6 h to 12 h post-reperfusion and reached its peak on 12 h post-reperfusion(P〈0.05).Urine VEGF significantly increased on 6 h post-reperfusion,whereas it significantly reduced on 24 h post-reperfusion(P〈0.01).Serum VEGF was positively correlated with urine levels of VEGF(P〈0.01).Conclusions Serum and urine VEGF level can be regarded as early monitoring indicator of ARF rat renal IRI.
