摘要 目的研究白细胞介素(IL)-1β和白细胞介素受体拮抗剂(IL-1ra)对结肠癌细胞表达uPA的影响,探讨IL-1β对结肠癌细胞迁移能力的影响。方法体外培养人结肠癌细胞,加入IL-1β和IL-1ra,应用半定量逆转录-聚合酶链反应方法,比较不同处理组结肠癌细胞中uPAmRNA的表达。结果IL-1β可促进结肠癌细胞的生长增殖,上调uPAmRNA的表达,呈一定的剂量效应关系;在时间系列组中,可检测到uPA mRNA的表达,以24 h表达最高。IL-1ra可以抑制IL-1β对uPA mRNA表达的上调作用。结论IL-1β促进结肠癌细胞生长增殖,刺激结肠癌细胞uPA高表达,从而促进结肠癌的迁移,IL-1ra可抑制IL-1β的作用。 Objective To study the effects of interleukin-1β(IL-1β) and interleukin-1 receptor antagonist(IL-1ra) on the expression of urokinase-type plasminogen activator(uPA) in colon carcinoma cells,and to explore the effect of IL-1β on the migration of colon carcinoma cells. Methods Colon carcinoma cells were cultured in vitro and treated with IL-1β and IL-1ra,the expression of uPA mRNA was examined by semi-quantitative RT-PCR. Results IL-1β could promote the proliferation of colon carcinoma cells and up-regulate the expression of uPA mRNA in a dose-dependent way.In the time series,the expression of uPA mRNA at 24 hours was the highest.The effect was inhibited by IL-1ra. Conclusion IL-1β promotes the proliferation and induces the expression of uPA in colon carcinoma cells.IL-1β can stimulate the migration of colon carcinoma cells.IL-1ra can block the action of IL-1β.
