摘要
目的在体外研究α2整合素受体与骨肉瘤细胞胞内基质金属蛋白酶2(MMP-2)的关系。方法用含不同浓度α2整合素单克隆抗体的DMEM培养基,培养MG63细胞6 h后,Trizol法提取细胞总RNA,RT-PCR半定量检测MMP-2的mRNA表达。分别用含浓度为10、25、50μg/ml的α2整合素单克隆抗体的DMEM培养基,培养MG63细胞48 h后,免疫细胞化学法检测MMP-2蛋白表达。结果随α2整合素单克隆抗体抗体浓度升高,MG63细胞MMP-2的mRNA和蛋白表达均降低。结论α2整合素受体可促进骨肉瘤MG63细胞MMP-2的表达,这种促进作用体现在RNA和蛋白水平。
Objective To study the relationship between cell surface integrin receptor(α2) and rnatrixrnetalloproteinases 2(MMP-2) of osteosarcoma in vivo. Methods After cultured respectively in DMEM media containing 10,25 and 50 mmol/L α2 integrin monoclonal antibody for 6 h, the RNA of MG63 cells was extracted using Trizol assay and mRNA was detected by semi-quantity RT-PCIL After cultured in DMEM media containing 10, 25 and 50 mmol/L α2 integrin monoclonal antibody respectively for 48 h, irnrnunohistochemisty assay was used to detect the MMP-2 protein expression. Results With the increasement of the concentrations of α2 integrin monoclonal antibody, the expressions of MG63 mRNA and protein were both down-regulated. Conclusion The α2 integrin acceptor on osteosarcoma cell surface may promote MMP-2 expression on the level of RNA and protein.
