摘要
目的:探讨阿司匹林作用下人子宫内膜腺癌Ishikawa细胞凋亡率与凋亡抑制蛋白Bcl-xl表达变化的关系。方法:应用流式细胞仪(FCM)检测不同浓度阿司匹林对子宫内膜腺癌Ishikawa细胞周期和凋亡率的影响;Westernblotting法检测阿司匹林对Bcl-xl蛋白表达的影响。结果:①阿司匹林引起细胞阻滞在G1期,细胞凋亡率增加,与药物作用浓度相关(P<0.05);②阿司匹林降低Bcl-xl蛋白的表达,与药物作用浓度相关(P<0.05);③Ishikawa细胞凋亡率与Bcl-xl蛋白的表达水平存在负相关关系(r=-0.873,P<0.05)。结论:Bcl-xl蛋白在阿司匹林促进子宫内膜腺癌细胞凋亡的调控中起到重要的作用。
Objective: To study the relationship between aspirin induced apoptosis rates and the expression of Bcl-xl protein in endometrial adenocarcinoma Ishikawa cells. Methods: Flow cytometry (FCM) was used to examine the distribution of cell cycles and the rates of cell apoptosis. Western blotting was used to evaluate the expression of Bcl-xl protein. Results: 1) Aspirin increased the distribution of G1 and the rates of cell apoptosis in a dose-dependent manner (P〈0.05). 2) Aspirin decreased the expression of Bcl-xl protein in a dose-dependent manner (P〈0.05). 3) The apoptosis rates of Ishikawa cells were negatively linearly related to the expression of Bcl- xl protein (r =- 0.873, P〈0.05). Conclusion: Bcl-xl protein may play an important role in the apoptosis of Ishikawa cells induced by aspirin.
