不同时机葛根素治疗对心肌缺血再灌注损伤的保护 被引量:5

Protective Effect of Puerarin Preconditioning on Myocardial against Ischemia-reperfusion Injury in Rats

摘要 目的:观察缺血前和再灌注前葛根素(PUE)治疗对大鼠心肌缺血-再灌注损伤(I/R)的保护作用。方法:开胸结扎左冠状动脉前降支(LAD)复制大鼠心肌I/R模型,设立假手术(sham)组、I/R组、PUE-A组(缺血前5min给药)和PUE-B组(再灌注前5min给药),比色法测定血清肌酸激酶(CK)活力,放射免疫法测定血浆内皮素(ET)浓度,硝酸还原酶法测定血清一氧化氮(NO)浓度,TTC法测定心肌梗死面积。结果:与I/R组比较,PUE-A、B两组血清CK活力明显降低,血浆ET浓度明显降低,血清NO浓度明显增高,心肌梗死面积明显缩小(P<0.05),且PUE-A、B两组间比较差异无显著性(P>0.05)。结论:缺血前和再灌注前葛根素治疗均能明显减轻大鼠心肌I/R损伤,且两种给药时机的保护效应相近。 Objective: To investigate the protective effect of puerarin (PUE) before ischemia or before reperfusion on myocard against ischemia-reperfusion injury in rats. Methods: Rats myocardial ischemia-reperfusion injury (I/R) were induced by ligating the left anterior descending coronary artery for 45 minutes and reperfusion for 120 minutes. Twenty-four rats were randomly divided into four groups (n =-6 each): sham-operation group (group S), ischemia-reperfusion group (group I/R), PUE treated groups (group PA received PUE at 100 mg/kg intravenously 5 minutes before ischemia, and group PB received PUE at 100 mg/kg intravenously 5 minutes before, reperfusion). Serum creatine kinase (CK) activity was measured with colorimetric method, concentrations of plasma endothelin (ET) detected with radioimmunoassay, and plasma NO content measured with Griess reaction assay. TTC dyeing was used to determine myocardial infarction sizes. Results: CK activity and ET concentrations were significantly lower, and the serum NO contents were significantly higher in groups PA and PB than in group I/R. Myocardial infarct sizes decreased dramatically in groups PA and PB, and there was no significant difference in all of the indexes between the 2 groups. Conclusion: Preconditioning with puerarin before ischemia or before reperfusion can reduce myocardial ischemia-reperfusion injury in rats, and the protective effects of puerarin injection at these 2 time points are similar.
