粉防己碱对阴茎海绵体平滑肌细胞抗氧化能力的影响 被引量:2

Antioxidant Effect of Tetrandrine on Cultured Rabbit Corpus Cavernosum Smooth Muscle Cells Injured by Hydrogen Peroxide

摘要 【目的】研究粉防己碱(Tet)对过氧化氢(H2O2)致新西兰白兔阴茎海绵体平滑肌细胞损伤的保护作用。【方法】体外培养新西兰白兔阴茎海绵体平滑肌细胞,随机分为3组:对照组、H2O2组和Tet+H2O2组。①四甲基偶氮唑盐还原实验观察Tet对H2O2诱导损伤的阴茎海绵体平滑肌细胞的存活率;②化学比色法观察Tet对H2O2诱导损伤的阴茎海绵体平滑肌细胞的乳酸脱氢酶(LDH)的释放、超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的影响。【结果】H2O2诱导阴茎海绵体平滑肌细胞4h,①细胞存活率由100%降至(48.57±4.1)%(n=8;P<0.01);②LDH释放由对照组的(487.3±78.6)U/L增加到(1105.5±65.8)U/L(n=8;P<0.01);③MDA含量由对照组的(3.8±1.4)nmol·L-1·mg-1增加到(77.3±8.6)nmol·L-1·mg-1(n=8;P<0.01);④SOD活性由对照组的(51.8±5.9)U/mL降为(30.7±4.5)/mL(n=8;P<0.01)。粉防己碱可浓度依赖性地提高细胞存活率;降低LDH释放和MDA含量,提高SOD活性。【结论】粉防己碱对H2O2致阴茎海绵体平滑肌细胞损伤具有保护作用,其作用机制可能与增强细胞抗氧化能力,减少脂质过氧化物导致的细胞膜损伤有关。 【Objective】 To investigate the antioxidant effects of tetradrine (Tet) on cultured corpus cavenosum smooth muscle cells (CCSMC) injured by hydrogen peroxide (H2O2). 【Methods】 The CCSMC were cultured in vitro and divided randomly into three groups: contrast group, H2O2 group, and Tet + H2O2 group. The effects of Tet on the viability of cell injured by H2O2 were assessed by 4,5-simethylthiazaoly colorimetric assay. The level of malondialdehyde (MDA), superoxide dismutase (SOD) activity and lactate dehydrogenase (LDH) release of CCSMC injured by H2O2 were detected by chemical colorimetry. 【Results】 After treatment with 1 mmol / L H2O2 for 4 h, the cell viability dropped from 100% to (48.57 ± 4.1)% (n = 8; P〈0.01). While LDH release increased from (487.3 ± 78.6) U / L to (1105.5 ± 65.8) U / L (n = 8; P〈0.01). The level of MDA increased from 3.8 ± 1.4 nmol·L^-1·mg^-1 to (77.3 ± 8.6) nmol·L^-1·mg^-1 (n = 8; P〈0.01) and the SOD activity dropped to (30.7 ± 4.5) U / mL from (51.8 ± 5.9) U / mL that in contrast group (n = 8; P〈0.01). However, treatment with different concentration of Tet (10^-7-10^-4 mol / L) could inhibit the damaging effects of H2O2, with increased cell viability and SOD activity (P〈0.01), and decreased LDH release and MDA content (P〈0.05 or P〈0.01), respectively. 【Conclusion】 Tet has protective effect on CCSMC injured by H2O2, which might be related to enhancing the antioxidant abilities of CCSMC, and decreasing the injury of cell membrane by lipid peroxidation.
