摘要
为了解狂犬病病毒(RV)的变异情况,本研究对广西不同地区的22个RV分离株的L基因3'端片段进行克隆和测序,与国内外发表的RV街毒、固定毒株及类RV株相应部分的多态性进行了比较分析。结果表明,所测定的22个广西RV野毒分离株属于基因Ⅰ型,可分为3个群即Ⅰ群、Ⅱ群和Ⅲ群。其中13株属于Ⅰ群,其核苷酸同源性为96.9%~100.0%,氨基酸同源性为98.2%~100.0%;8株属于Ⅱ群,株核苷酸同源性为96.5%~99.7%,氨基酸同源性为97.8%~100.0%。仅1株属于Ⅲ群。22个RV分离株与RV固定毒株核苷酸和氨基酸的同源性分别为79.5%~86.9%和85.8%~96.5%,与类RV核苷酸和氨基酸的同源性分别为65.7%~70.3%和70.4%~76.5%。
In this study the cDNA sequences of L gene 3′ terminal of twenty-two rabies virus (RV) isolates from different regions of Guangxi province were cloned and sequenced. The nucleotide sequences and deduced amino acid sequences of the L gene 3′ terminal region of these isolates were compared with that of fixed virus strains and rabies-related virus strains published previously. The results indicated all RV isolates from Guangxi were genotype Ⅰ of Lyssavirus and could be divided into three groups as Ⅰ, Ⅱ and Ⅲ. Of which, thirteen isolates were classified to group Ⅰ with a identity of 96.9 % to 100 % at nucleotide level (NL) and 98.2 % to 100 % at amino acid level (AAL); Eight isolates belonged to group Ⅱ with 96.5% to 99.7% at NL and 97.8 % to 100 % at AAL; Only one isolate belonged to group Ⅲ. Comparing the isolates with the fixed RV strains, the identity were 79.5 % to 86.9 % at NL and 85.8 % to 96.5 % at AAL, respectively, and the homologies were 65.7 % to 70.3 % at NL and 70.4 % to 76.5 % at AAL, respectively, compared with rabies-related virus strains.
