基因工程抗菌肽表达条件的优化及生物活性试验 被引量:4

摘要 探讨表达条件对重组抗菌肽表达量和抗菌活性的影响。选用SMD1168毕赤酵母菌株表达cecropinB及其串联体基因和cecA-mag(cecropin A前8个碱基和magainin的第9个碱基)的3个突变体基因抗菌肽。对6种重组酵母菌的摇瓶发酵条件,如pH、发酵温度、甲醇的添加量等进行了优化,结果优化后的表达条件可提高重组抗菌肽的表达量和抗菌活性。并从抑菌试验、酸碱稳定性试验、热稳定性试验、体外稳定性试验探讨了其生物学活性。 To study the effect on expression quantity and antibacterial activity of the ABP's recombinant Pichia pastoris,six recombinant antibacterial peptides,cecroin B,ceropin B-3,cecA-mag and three mutants of cecA-mag were studied.The requirements for the flask-shaking culture fermentation,such as pH,culture temperature and so on,were optimized.It was found the expressed level of the recombinant antibacterial peptides,could be increased.Biological activity was studied by agarose diffusion assay,heat-stable and acid-stable experiments.Different culture methods have effect on expression level and antibacterial activity.
