摘要
将流感病毒分别接种于鸡胚成纤维细胞(CEF)、Vero和MDCK细胞,并改变细胞维持液中FBS浓度,然后检测胰酶的用量、细胞培养流感病毒的最佳时间、细胞接种病毒最佳吸附时间和接种剂量、收获细胞液后的血凝素(HA)滴度。结果表明,胎牛血清对病毒的繁殖有明显抑制作用;加入约4μg/mL的胰酶可提高病毒产量。最佳收获时间为60 h^72 h,吸附时间为90 min,接种剂量为0.1 mL/L;CEF、MD-CK细胞上病毒HA滴度高于Vero细胞。
The FBS concentrations in sustaining cell cultures were changed.Then,The dose of trypsin,the optimal time for viral culture,the optimal viral obsorption time and inoculation dose,and the HA titers of influenza virus were tested.The results indicated that FBS inhibited obviously the proliferation of influenza virus.4 μg/mL trypsin increased the viral output.The optimal harvest time was 60-72 h,the absorption time was 90 min,and the inoculation dose was 0.1 mL/L.The viral HA titers in CEF and MDCK cells were higher than that in Vero cells.
