姜黄素抑制乳腺癌MCF-7细胞增殖及其相关的氧化应激机制 被引量:13

摘要 目的:探讨姜黄素抑制乳腺癌MCF-7细胞增殖及其相关的氧化应激机制。方法:用不同浓度(5~40μmol/L)的姜黄素作用于乳腺癌细胞株MCF-7细胞,以MTT法检测在6、12、24、48h的细胞增殖率,流式细胞术检测细胞凋亡率,DCFH-DA染色流式检测细胞内活性氧(reactive oxygen species ROS)生成变化,黄嘌呤氧化酶法检测氧化物歧化酶(superoxide dismutase,SOD),可见光分光光度计法检测过氧化氢酶(catalase,CAT)活性。结果:低浓度(5、10μmol/L)姜黄素作用后MCF-7细胞增殖受抑制(P<0.01),但无凋亡发生;伴随细胞内ROS短暂升高后随时间逐步下降,SOD、CAT先下降后逐步提高。高浓度(20、40μmol/L)姜黄素作用后细胞增殖抑制率明显增加(P<0.01),细胞出现大量凋亡;伴随细胞内ROS即刻明显升高,随时间延长无明显下降;SOD、CAT明显下降(P<0.01)。结论:姜黄素具有剂量依赖的抗氧化与促氧化双重作用,不同浓度的姜黄素通过对乳腺癌细胞内氧化还原状态的调节参与了其抗肿瘤细胞增殖作用的机制。 Objective:To investigate the inhibitory effect of curcumine on the proliferation of breast cancer MCF7 cells and the related oxidative stress mechanism. Methods: Human breast cancer MCF7 cells were treated with different concentrations of curcumine (540 μmol/L), then the proliferation of MCF7 cells was examined by MTT assay after 6, 12, 24 and 48 h; the apoptosis rate of MCF7 cells was detected by flow cytometry (FCM); the levels of reactive oxygen species (ROS) in MCF7 cells were detected by DCFHDA staining; and the activities of superoxide dismutase (SOD) and catalase (CAT) in MCF7 cells were detected by xanthine oxidase assay and visible spectrophotometer, respectively. Results: Low level curcumine (5, 10 μmol/L) inhibited the proliferation of MCF7 cells (P〈0.01) without inducing apoptosis; ROS levels in MCF7 cells were firstly increased and then declined gradually; meanwhile, SOD and CAT activities decreased initially and then gradually increased. High level curcumine (20, 40 μmol/L) significantly inhibited the proliferation of MCF7 cells and induced evident apoptosis (P〈0.01), and ROS levels in MCF7 cells were elevated instantly and maintained for a long time after treatment; at the same time, SOD and CAT activities were significantly decreased (P〈0.01). Conclusion: Curcumine has a double role of antioxidant and oxidation, and it functions in a dosedependent manner. Different concentrations of curcumine can inhibit the proliferation of tumor cells through regulating oxidative statuses in cells.
