摘要
目的观察艾拉莫德对小鼠成纤维细胞3T6细胞分泌胶原的影响。方法利用MTT法得到艾拉莫德对3T6细胞的半数毒性浓度约为1142.6mg/L,最终给药浓度选取334.38mg/L(二甲基亚砜浓度为1μl/256μl)、167.4mg/L、83.7mg/L、41.85mg/L,并以未加药组作为空白对照,分别在给药后12h、24h、36h、48h取细胞上清,采用氯胺T法检测其中细胞分泌的胶原。结果药物浓度334.38mg/L组在给药后12h、24h、36h与空白对照组比较胶原浓度增大(P<0.05),167.4mg/L和83.7mg/L组在给药后24h、48h与空白对照组比较胶原浓度增大;41.85mg/L组在给药后12h、24h与空白对照组比较胶原浓度降低(P<0.05)。结论艾拉莫德对小鼠成纤维细胞3T6细胞分泌胶原的影响具有时间和药物浓度依赖性,高浓度可促进胶原的分泌,低浓度药物41.85 mg/L组可减少细胞胶原的分泌。
Objective To study the effect of Iguratimod on the collagen secretion of 3T6 cells. Method The median toxic concerttration of Iguratimod was determination by MTT Assay. Iguratimod' s median toxic concentration to 3T6 cell was about 1142.6mg/L. Taking into the consideration of the cells' performance after administration under spectacles, we chosen the administration concentration as 334.8 mg/L( DMSO : 1 μl/256μl) , 167.4mg/L,83.7mg/L and 41.85 mg/L. After administration of 12 hours, 24 hours, 36 hours and 48 hours, the supernatant was collected for assayed respectively. The collagens content was detected by the method of chloramine T. Results The collagen concentration of the drug concentration 334. 375mg/L group increased (P 〈 0.05) after 12 hours, 24 hours and 36 hours of administration in comparison with the normal blank group. After 48 hours, their collagen concentration decreased. The collagen concentration of group 167.4mg/L and group 83.7mg/L increased after 24 hours and 48 hours of administration in comparison with the normal blank group. The collagen concentration of group 41.85mg/L decreased after 12 hours and 24 hours of administration in comparison with the normal blank group(P 〈 0.05). Conclusion The effect of Iguratimod on the collagen secretion of 3T6 cells has a time - dependence and dose -dependence. The high concentration can promote the collagen secretion. The low concentration 41.85mg/L can reduce the col- lagen secretion.
