中药安胎养血合剂干预肾移植大鼠肾细胞凋亡及Fas/FasL基因的表达(英文) 被引量:1

摘要 背景:前期实验表明,作者自制的中药复方安胎养血合剂能够抑制肾移植大鼠急性排斥反应,延长受体存活时间。目的:观察安胎养血合剂对异基因肾移植大鼠移植肾肾小管上皮细胞凋亡及Fas/FasL基因表达的影响,并与同基因肾移植大鼠进行对比。设计、时间及地点:随机对照动物实验,于2005-07/2006-06在辽宁医学院附属第一医院外科实验室完成。材料:选用雄性SD大鼠24只,雄性Wistar大鼠48只。方法:按体质量相近配对后,按随机数字表法分为3组,每组12对。对照组大鼠同基因肾移植(Wistar→Wistar大鼠),生理盐水灌胃;生理盐水组大鼠异种基因肾移植(SD→Wistar大鼠),肾移植后不用任何免疫抑制剂,生理盐水灌胃;安胎养血合剂组大鼠异种基因肾移植(SD→Wistar大鼠),肾移植后给安胎养血合剂灌胃,灌胃剂量均为10mL/(kg·d),移植后1次/d。主要观察指标:于移植后第5天处死动物取肾脏组织,通过原位末端标记法检测凋亡指数,采用反转录-聚合酶链反应检测Fas/FasLmRNA表达。结果:生理盐水组移植肾组织凋亡细胞最多,对照组凋亡细胞极少见。生理盐水组和安胎养血合剂组凋亡指数显著高于对照组(P<0.05),安胎养血合剂组凋亡指数显著低于生理盐水组(P<0.05)。各组中均有FasmRNA的表达,FasLmRNA的表达水平在安胎养血合剂组中明显低于生理盐水组(P<0.05)。结论:安胎养血合剂可以通过下调大鼠移植肾肾小管上皮细胞FasL的mRNA表达,从而减少移植肾的细胞凋亡,起到免疫抑制剂的作用。 BACKGROUND:Our previous primary study had approved that traditional Chinese medicine Antai Yangxue mixture might resist acute rejection and prolong rat transplanted kidney survival.OBJECTIVE:To study the effect of Chinese herbal medicine Antai Yangxue mixture on renal tubule epithelial cell apoptosis and Fas/FasL gene expression in rat allogenic transplanted kidney tissue,and to compare with syngeneic transplanted kidney rats.DESIGN,TIME AND SETTING:The randomized,controlled,animal experiments was performed between July 2005 and June 2006 at the Laboratory of Department of Surgery,First Affiliated Hospital,Liaoning Medical University,Jinzhou,Liaoning Province,China.MATERIALS:A total of 24 male Sprague Dawley rats and 48 male Wistar rats were used for this study.METHODS:Following matching according to body mass,rats kidney transplantation models were established,and they were divided into control group(Wistar→Wistar rat),saline group(Sprague Dawley,SD→Wistar rat)and Antai Yangxue mixture group(SD→Wistar rat).Control group and saline group were administered with saline through lavage.Rats in the Antai Yangxue mixture group underwent Antai Yangxue mixture following renal transplantation,10 mL/kg per day,once a day.MAIN OUTCOME MEASURES:The animals were sacrificed at day 5 after transplantation to obtain kidney tissues.Apoptotic index was measured by in situ end labeling.Fas/FasL mRNA expression was detected by RT-PCR.RESULTS:The number of apoptosis cells in saline group was the largest,and that of the control group was smaller.Apoptotic index was significantly greater in the Antai Yangxue mixture group and saline group compared with the control group(P〈0.05).Apoptotic index was significantly lower in the Antai Yangxue mixture group than in the saline group(P〈0.05).Fas mRNA ex-pression could be detected in each group.FasL mRNA expression was significantly lower in the Antai Yangxue mixture group than in the saline group(P〈0.05).CONCLUSION:By decreasing the expression of FasL mRNA,Antai Yangxue mixture can reduce apoptosis of transplanted kidney and play effects as immunosuppressive agent in renal tubular epithelial cells of rat transplanted kidney.
