摘要 目的:评估脂质体介导环氧合酶(COX)-2反义寡核苷酸(AsODN)对皮肤鳞状细胞癌细胞株Colo-16生长及血管内皮生长因子(VEGF)表达的影响。方法:将COX-2无义寡核苷酸(NsODN)和AsODN用脂质体分刖导入Colo-16细胞中,克隆并观察COX-2 AsODN对细胞的生长抑制作用。Hoechst33258荧光染色法和流式细胞仪观察Colo-16细胞凋亡形态学变化及凋亡率。Western blot检测VEGF表达水平。结果:COX-2 AsODN作用于Colo-16细胞后,增殖能力受到抑制,克隆形成率下降(P<0.05),细胞出现明显凋亡,COX-2 AsODN组细胞早期凋亡率显著高于对照组(P<0.05);VEGF表达明显下调结论:COX-2AsODN能够抑制Colo-16细胞体外生长,诱导其凋亡,并能显著下调VEGF表达。 Objective: To investigate the effects of COX - 2 AsODN on Colo - 16 cells growth and the expression of VEGF gene in Colo- 16 cells. Methods: The nonsense and antisense oligonucleotides targeting cyclooxygenase- 2 were respectively transfected into Colo- 16 by liposome. The growth proliferation of Colo- 16 cell was estimated by colony assay. The morphological change of apoptotic cells and apoptosis rate were tested by Hoechst 33258 fluorescent staining and flow cytometry respectively. The protein expression of VEGF was detected by Western blot. Results: After the transfection with antisense oligonucleotides targeting cyclooxygenase - 2, the growth of Colo - 16 cells was obviously inhibited and the number of cell colony was reduced ( P 〈 0. 05 ). The morphology of apoptosis was observed in Colo - 16 cells. The early apoptosis rate was higher than that in control group ( P 〈 0.05). The expression of VEGF was downregulated. Conclusion: Antisense oligonucleotides targeting cyclooxygenase- 2 can inhibit the growth and accelerate apoptosis of Colo - 16 cells, and reduce VEGF gene expression in Colo - 16 cells.
